The
analysis of results of experimental and morphological investigation of
supraperiostal and subperiosteal osteoplastic materials injecting(PRP and
Biomin GTLS)
The
urgency of the problem. In
modern periodontology widely used technologies, which contribute the
optizimation of regeneration processes in peridontal tissues. More
frequently appears the results of
efficiency of platelet-rich plasma(PRP)
use in dentistry, particularly if it combinates with osteoconductive material.
The object of study:To
track the morphological changes, developed in peridontal tissues in experiment
on rats, using local isolate and combined injection of PRP and osteoplastic material Biomin GtlS.
The material and methods
of study. Experimental and morphological fragment of investigation carried out on 30 rats on the
base of vivarium of state institution “Crimea state medical
university named after S. I. Georgievsky” .Were formed 2 experimental groups of
investigation ,15 animals in each group. In first group performed once
occurring subperiosteal injection of PRP. In second group performed injection of PRP with osteoplastic material Biomin GtlS by same
method.
The blood sampling
performed by disposable syringe from the vessels of tail of rat. Heparin was
injected into the syringe(0,05 U). The taken blood was centrifuged within 10 minutes at 2000 r/min within 15 minutes at
6000 r/min. From centrifugate
selected the lower part of plasma,
yellow – pink color(0,1-0,15 ml). The received PRP (0,1 ml) injected by insulin
syringe in supraperiostal ans subperiosteal molar area of rats lower jaw.
In second experimental
group of animals injected Biomin
GtlS(0,1gr.) with PRP(0,1 ml) in the same area.
Euthanasia of the rats carried out by decollation during the ether
anesthesia on 7th, 10th, 30th and 60th
day(3 individuals). The biopsy for following microscopic examination prepared
by common pathohistological methods.
There are studied 60
preparations, received from 30 rats in both groups.
Results of
study. Our suggested
method of osteoplastic material
injection allows to use the least traumatic way of target delivery,
including point delivery, by
subperiosteal location of preparations. This way increased the ability
of amount of administered drug dosage
and it preservation in the area of injection without significant migration and premature washout by the blood. The
fibrin clot formation contributes the fixation of the osteoplastic material in
tissues, that improves condition for
retention of particles of
Biomin GtlS. The powder Biomin allows
to use small diameter needles. It decreases trauma of the procedure and
the severity of inflammation.
In this way, in early
postinjective period the local signs of inflammation (edema and hyperaemia) in
groups of comparison visually were
expressed not significantly and
subside on 1-3(hyperaemia) and on 3-5(edema) day of observation with not
significant intergroup differences. The signs of postinjective inflammation
processes formation. After attenuation of the inflammatory process in area of
osteoplastic materials injection visualy and by palpation noticed the growth of
tissues, which was more exspressive in
second group.
The results of histological
investigation allowed to determine , that the local and dystrophic changes were
expressed equally moderately. Tissues were platelet-impregnated.The
proliferative reactions traced in epithelial and submucous layers, in
periosteum, in bone and even in peridont. On 10th and 20th day of
experiment determined the signs of the damaged tissue structures recovery and
the formation of new structures, in the form fibroblasts growth, connective tissue, the appearance of
osteoblasts, the formation of new blood vessels and structures, reminding haversian canals and trabecula of bone.
These signs testify about active osteoinductive action of osteoplastic
materials combination.On 30 and 60 day histologically noticed the completion of
the recovery processes by newly formed coarse-fibered connective tissue forming, included trabeculae of bone and septa,
a lot of osteoblasts, structure along the osteon and haversian canals.
Conclusion. During
the study of clinical and morphological changes , developed in peridontal
tissues in experiment on rats using PRP and osteoplastic material Biomin GtlS
were traced the signs of recovery processes optimization. In our opinion, this
is due to the complementary properties of these preparations, which control the
local regeneration processed in peridontal tissues. Injection of PRP forms the
high concentration of growth factors at the injection site , PRP is biostimulant
and demonstrates immunocorrective, anti-inflammatory and osteoinductive
effects. Biomin GtlS has osteoinductive
effect, contains argent, which has local antimicrobial and anti-inflammatory
effects.
