Influence bridges based on implants on
cytochemical indicators on the state of peripheral blood neutrophils
In dental implantology
despite of significant progress and scientific achievements, as before, remain
relevant issues related to the prevention of inflammatory complications in
tissue complex prosthetic support zones.
Cytochemical studies were used to assess the activity of dehydrogenases
of peripheral blood neutrophils, activity of LDH and LDH.The data obtained in
the study of the treatment of orthopedic patients with bridges relying on
implants can be the basis for the possibility of reducing the likelihood of
complications.The aim of our study was to conduct monitoring cytochemical
peripheral blood neutrophils in orthopedic patients after installing bridges
implant.
Methods of investigation
Pictures of our study served as peripheral blood of patients who
underwent dental implantation . To conduct the study were examined 26 people of
both sexes , requiring orthopedic treatment with bridges .Cytochemical methods
of research dehydrogenase activity in peripheral blood neutrophils .Certain
interest to study the activity of dehydrogenase enzymes of glycolysis and the
Krebs cycle . Quadrupeds that succinate dehydrogenase and lactate dehydrogenase
is one of the most important cellular enzymes.
Dehydrogenase - enzymes
that cleave hydrogen from an appropriate substrate and transfer it to the
acceptor . This mechanism of action used in cytochemical methods , which are
introduced into the system indicator assumes hydrogen. The indicator thus
should be of a color and precipitate. For these purposes, the most commonly
used water-soluble tetrazolium salt : neotetrazolny chloride ( NT ), blue
tetrazolium ( PT) nitrotetrazolny blue (NBT ) and others that under the action
of reducing agents are converted into water-soluble colored formazan , allowing
you to install intracellular localization of dehydrogenases under a light
microscope.
Method for studying the
activity of dehydrogenases in blood neutrophils proposed by us when cells of
heparinized blood smears made after treatment with appropriate
reagents hour after incubation.In studies as an indicator enzyme process used
nitgrogetrazoly blue ( NTS ) , which forms the reduction in cell fine granules
of formazan staining cytoplasm of gray to deep blue. We used native thin blood
smears , air-dried , which, after appropriate processing . Incubated for 45
minutes at 37 ° C. Cell nuclei dokrashivali solution Methylene green . Dried
smears microscope with immersion lens microscope MBI- 15.To determine the
activity of LDH apply solutions : sodium cyanide , sodium lactate , NST .
gemodez , NAD . Deposition granules formazan observed at the location of EDC .
Staining intensity ranged from smoky gray to intense blue , cell nuclei were
well circumscribed .To determine the activity of LDH - solutions : NBT ,
succinate Katra , phosphate buffer . There was a clear localization of formazan
granules . The activity of LDH was judged by the intensity of formazan
deposits granules . To assess the activity of enzymes in blood cells , we
calculated the average cytochemical ( SCP ) .
Cytochemical study of
peripheral blood neutrophils .In the analysis of the cytochemical activity of
peripheral blood neutrophils in patients with orthopedic established with
bridges revealed :By 1 month after prosthesis wearing light and insignificant
character ( P> 0.05 ) , enzyme imbalance , so the activity of succinate
dehydrogenase ( LDH ) were 1 , B7 ± 0,06 conv , which was 9.2% below control
and enzymatic activity of lactate dehydrogenase (LDH ) 2,48 ± 0,16 conv that
was by 7.8 % higher than control Are .To 3 months after prosthesis with bridges
imbalance decreased since LDH activity was 1,72 ± 0,09 conv , which was 6.8%
below koshrolnyh indicators, and LDH activity was 2,41 ± 0,11 conv. u , which
was 4.7% higher than the control ( P > 0.05).To 6 - 12month , we were treated
as long-term results , cytochemical activity of peripheral blood did not differ
from controls.
Conclusions
Detailed analysis of cytobiochemicical studies allowed to conclude that
the installation of bridges (made of acrylates ) implant has no significant
changes in the state of neutrophils in the peripheral blood of patients , as
evidenced by indicators dehydrogenase activity of peripheral blood neutrophils
and LDH and LDH .
