THE EFFECT OF HYPОCALLORIC DIET ON MORPHOLOGY OF THE EXOCRINE PANCREAS IN ONE-MONTH OLD OFFSPRING OF RATS

Introduction. Damage of the pancreas caused by an unbalanced diet, is an urgent problem of medicine and currently requires the attention of researchers is the actual medical problem [1, 2]. The urgency of this direction for Pediatric Gastroenterology determined, pregnant women are increasingly consume hypocaloric diet with protein deficiency, on the other hand the fact that the negative effects of such a diet on pancreatic structure of their offspring are still poorly understood [3].

The aim of the study was to investigate the morphological changes of the exocrine pancreas in rats from mothers who received hypocaloric diet.

Object and methods. The morphological changes of the pancreas one-month rats. The main group consisted of 1-month-old rats (10 animals) were born to mothers who received hypocaloric diet [4] during pregnancy (gr. 1). The comparison group (gr. 2) were rats of the same age (10 goals) from rat mothers who received a balanced diet.

Morphological examination included histological, morphometric, histochemical methods. We used routine staining methods: pikrofuksin Van Gisone, with hematoxylin eosin, methods by Mallory, methods by Brachet reaction CHIC and Hale, methods by Feulgen-Rossenbeka.

Photographing micropreparations and morphometric study was carried out on the microscope Olympus BX-41 using programs Olympus DP-Soft (Version 3: 1).

Statistical analysis was performed using Microsoft Excel-2003, Biostat.exe-2008 software programs using one-way ANOVA.

The results of research. Microscopic pancreas has lobed structure, connective layer between the hardware segments, with ample signs of the immaturity of the connective tissue, and edema (at 100% rats) - see Figure 1.

of parenchymal component is 64,7 ± 2,1 (p <0.001) of the total prostate volume (control 73,4 ± 1,9). Atrophy of the parenchyma with the growth of substitution of connective tissue found in all cases. 50% ± 15,8 (p <0.05) observations indicated focal lipomatosis.

At 100% of the rats exocrine main group with hydropic changes in the cytoplasm. The nuclei when stained by Feulgen-Rossenbeka weak (+ - ++) purple. There endocrinocytes with degenerative changes of the nuclei in the form karyopyknosis, krioreksisa and karyolysis (at 100%). At coloring by Brachet marked decrease (+ - ++) cytoplasmic staining intensity. Also decreased (+) PAS-positive cytoplasmic staining reaction when setting CHIC + Hale. This shows a decline in protein synthesis and mucus in the exocrine pancreas department.

According to the morphometric study the relative volume of the stromal component of (35,3 ± 2,1)% of total prostate volume (see. Figure 2). The stroma of the prostate in 100% of cases, symptoms of edema and focal sclerosis determined, 50% of focal lipomatosis. The vascular network is represented by RV and Lobular interlobular blood vessels, which have a well-defined clearance and are characterized by moderate congestion. Intralobular and interlobular ducts of the pancreas with a fine PAS-positive wall wide lumen, lined with high prismatic epithelium, degenerative changes which are expressed mild.

Conclusions:

1. The animals of the main group in terms of termination after the birth of hunger exocrine pancreas device undergoes a series of morphological changes. Revealed atrophy of the exocrine parenchyma, increased stromal component, degeneration of the cytoplasm and nucleus.

2. The results of the study indicate that hypocaloric diet of pregnant rats a significant risk factor for development of chronic pancreatic insufficiency later in their offspring.

ВЛИЯНИЕ ГИПОКАЛОРИЙНОЙ ДИЕТЫ НА МОРФОЛОГИЧЕСКОЕ СОСТОЯНИЕ ЭКЗОКРИННОЙ ЧАСТИ ПОДЖЕЛУДОЧНОЙ ЖЕЛЕЗЫ ОДНОМЕСЯЧНОГО ПОТОМСТВА КРЫС

В поджелудочной железе одномесячных крысят, рожденных от крыс получавших гипокалорийную диету в период беременности, экзокринный аппарат поджелудочной железы претерпевает ряд морфологических изменений. Выявлена атрофия паренхимы, увеличене стромального компонента, дистрофия цитоплазмы и ядер, что  является существенным фактором риска развития панкреатической недостаточности.

Вплив гіпокалорійної дієти на морфологічний стан екзокринної частини підшлункової залози одномісячного потомства щурів

У підшлунковій залозі одномісячних щурят, народжених від щурів, які отримували гіпокалорійну дієту в період вагітності, екзокринний апарат підшлункової залози зазнає ряд морфологічних змін. Виявлена атрофія екзокринної паренхіми, збільшення строми, дистрофія цитоплазми і ядер, що є фактором ризику розвитку панкреатичної недостатності.
