THE DISK WITH SPHERICAL MOVEMENT FOR MASS-EXCHANGE

 

The utility model concerns to the biotechnology and can be used for the cultivation of cells in the manufacture of viral vaccines (from Latin vaccine), and also in the variety of biologically active preparations.

The known design of the device for the cultivation of cells (AC) which contains the cylindrical case with technological branch pipes and a mixing disk from not magnetic material cantileverly fixed at the bottom of the case, and also a drive in the form of located on a disk and a rotor of magnets periodically co-operating among themselves (À.c. The USSR ¹1306944, C12 M3/00, 1987).

The lack of this device consists in low efficiency of hashing that is a consequence of console fastening of the disk. This feature convincingly explains the reason of decline of productivity of work of the device.

The device accepted for a prototype which contains the cylindrical case with the bottom from not magnetic material and the technological branch pipes, the established disk executed from a magnetic material at the bottom of case with the central core on its one part, and also a drive of a disk in the form of in regular intervals located on a circle under the case bottom consistently connected to the power supply of electromagnets, is the closest to the utility model for the technical essence and the achievable effect. The lack of this device consists in its low efficiency of hashing which inevitably generates decline of productivity of the cultivation and assists development of other lacks.

The denoted lack is caused by essentially passive mixing of cultural liquid in its top and average layers as the disk located on the bottom of the case has no elements directly co-operating with them and does not serve as the reason of their movement.

In a basis of useful model the problem of improvement of the device by disk maintenance additional mixing element is put. It should cause more active movement of cultural liquid in its top and average layers which will inevitably assist intensity and uniformity of the mass exchange. Such technical decision will lead to growth of productivity of work of the device as a whole.

The outlined problem is solving by that in the device for the cultivation of cells which contains the cylindrical case with the bottom of non magnetic material and technological branch pipes both placed at the bottom of the case and the disk executed from a magnetic material with the central core on one part, and also a drive in the form of  regular intervals located on a circle under a case bottom consistently connected to the power supply of electromagnets, according to the utility model the new there is that the disk is equipped by an additional central core which is located on an opposite side of a disk.

The disk equipment with an additional central core which is located on its other side intensifies movement of the top and average layers of cultural liquids which are raised by uniformity of mass exchange owing to what practical bases of acceleration of growth of cages are created. Thus, productivity of the cultivation rises promptly.

On the fig 1 the offered apparatus for the cultivation of the cells is represented schematically.

The apparatus consists of the cylindrical case 1 with the bottom 2 made of the non-magnetic material and the cover 3 with the technological sockets 4 for the removal of the cultural broth.

At the bottom 2 of the case 1 there is a mixing disk 5, made of a non-magnetic material with a central rod 6. At the opposite side of the disk 5 the central rod 7 is fixed. This rod 7 has to be exceeded by the high the level of the cultural broth 8 in the case 1 and may be carried out combined with the rod 6 as the one whole pressed in the disk 5 detail (not shown). The driving of the disk 5 is realized due to the electromagnets 9, which is located evenly on the circle under the bottom 2, which is connected consistently to the power supply 10 in the impulsive programming regime.

 

The device for the cultivation of cells works in the following way. With the engaging of an electromagnetic drive the electromagnets 9 make a rotating magnetic field which draws a disk 5 owing to what it comes to spherical movement round the bottom centre. Under these conditions, a disk 5 and a core 7 carry out directed moving of the cultural liquid 8, a disk 5 - mainly benthonic layer, and a core 7 - the top and average layers.

In comparing with a prototype, the offered device provides the improvement of quality of the mass exchange and, naturally, to uniformity of distribution of nutrients and cells in all volume which assists to an active growth of cells, by the expense of additional mixing of cultural liquid by a core 7. These signs serve as the underground to the growth of productivity of work of installation as a whole.

Decrease of the virulence of the microorganisms under the condition of preservation initial antigenicity and immunogenicity are the major principle of reception of the living vaccines.

Thus, in a basis of the way of the reception of living vaccines are laying the directed cultivation of the microorganisms on the nutrient mediums.

Live vaccines, modulating the immune answer adequate to the transferred disease, have essential advantages before other vaccines to this indicator.
