APOPTOSIS MARKER INDICATORS APO2.7 AND CD95

 AS DIAGNOSTIC CRITERION OF ADAPTATION IN CEREBRAL PALSY CHILDREN WITH DYSADAPTATION SYNDROME

Adaptation on the resort – an adaptation process to new environmental conditions, which occur in treatment and rest. There are features of adaptation processes in children, protracted and often ill, who arrived in a spa treatment at health resorts of Crimea from Ukraine. It is known that the degree of stress on physiological systems is caused by a contrast between climatic conditions and location. In particular category of children emergence of stress that can affect the immune system is manifested by sharp breakdown of adaptation. The aim of our study was to investigate the immune status in children with the dysadaptation syndrome, as well as monitoring of cellular immunity by apoptosis markers CD95 and APO2.7. We have examined 40 children of both sexes aged 7 to 15 years arrived on the resort stage of rehabilitation in the sanatorium "Anniversary" and "Emerald" in Evpatoria and treated as long-term and often ill, mainly chronic diseases of nazo-farynheal region. Assessment of adaptation in children lasted for category long and often ill included analysis of peripheral blood of patients with using of following markers: CD95 - cell surface receptor that can trigger apoptosis in it after its interaction with Fas-ligand or monoclonal antibodies to Fas, APO2.7 - for percent cells determination had undergone apoptosis. Method of assessment of adaptation using markers CD95 and APO2.7 includes using of flow cytometry and marker-receptor CD-95, as indicator pf apoptosis. Additionally we used a APO2.7 marker antigen (also known as 7A6), which appears on the membrane of mitochondria the early stages of apoptosis. APO2.7 expression appears in the early stages of apoptosis. APO2.7 also can occur in cells after irradiation, drugs or binding of CD-95 with Fas-ligand. Living cells are negative to this antigen. Analysis of the data indicates that increased apoptosis in  children at an early stage of adaptation deviation from the norm (increasing CD95 expression and APO2.7, indicating the development processes of conflicts). The degree of degradation of cellular immunity is reflected in the opposite correlations between cellular immunity (reduction of CD3 + and CD4 +) and apoptotic indices (increase of CD95 and APO2.7) from the analysis of the data suggests that children with deviations from the norm of approximation by identifying markers of peripheral blood neutrophils apoptosis occurred enhance apoptosis. We paid attention to the close of feedback (r> -0,5) between indicators of cellular immunity (reduction of CD3 +, CD4 +) and indices of apoptosis (increasing level of CD95 and APO 2.7) during exploring correlations between the indices of cellular immunity indexes and apoptosis. Thus, the anomaly of approximation by measurement of apoptosis of peripheral blood neutrophils was found in a clear dependence on the medical activities at the stage of sanatorium and resort rehabilitation.
